Description:
Transgenic line overexpressing GLK2. The genomic sequence of GLK2 was amplified from genomic DNA and cloned into pCR8/GW/TOPO to generate an entry clone. The GLK2 entry clone was recombined into the pGWB405 destination binary vector for C-terminal fusion with GFP and expression driven by the CaMV 35S promoter. Col-0 plants were transformed by the floral dip method and selection was performed based on resistance to kanamycin. Homozygous for the transgene.

Dark-green leaves, reduced shoot growth, late flowering, enhanced chloroplast production in the roots. Greater guard cell chloroplast area, and a slight increase in size of pavement cell chloroplasts with a slight trend toward increased pavement cell chloroplast numbers. Increased NPQ (nonphotochemical energy quenching) and a decrease in the flux of electrons through PSII.
