Description:
Transgenic line overexpressing GNC in the glk1 glk2 double mutant background. The genomic sequence of GNC was amplified from Col-0 genomic DNA and cloned into pCR8/GW/TOPO to generate an entry clone. The GNC entry clone was recombined into the pGWB20 destination binary vector for C-terminal fusion with a 10x Myc tag and expression driven by the CaMV 35S promoter. glk1 glk2 double mutant plants (generated by genetic crossing of CS9805 to CS9806) were transformed by the floral dip method and selection was performed based on resistance to kanamycin. Homozygous for the transgene.

Partial rescue of the glk1 glk2 mutant phenotype. Small rosette, light-green leaves. Reduced sizes of mesophyll chloroplasts. Enhanced chloroplast production in both epidermal pavement and stomata guard cells, as well as in roots and hypocotyls. Increase in NPQ (nonphotochemical energy quenching) and a decrease in the flux of electrons through PSII.
