Description:
Transgenic expression of a kinase-inactive mutant of ETR1 (tETR1-G2) in the etr1-9;ers1-3 double mutant background. An ETR1 genomic clone containing the promoter and full-length coding regions was subcloned into pALTERII and the Altered Sites Mutagenesis System was used for site-directed mutation of the G2-box of ETR1. This mutation codes for a conversion of Gly-545 (GGG) to Ala (GCG) and a conversion of Gly-547 (GGC) to Ala (GCC). The mutant product was then cloned into binary vector pCAMBIA2380. Since the etr1-9;ers1-3 homozygous double mutant is sterile, the construct was initially transformed into etr1-9/etr1-9;ers1-3/ERS1 plants, and plants homozygous for the mutations and the transgene were identified in subsequent generations. Kanamycin resistant.
