Description:
elf3-14 mutant identified from a forward genetic screen for PRR7 protein turnover factors. EMS mutagenesis was performed on a transgenic line expressing a PRR7::PRR7-luciferase translational fusion. To generate the transgenic line, the PRR7 coding sequence was subcloned into a pPZP-BAR DONR plasmid harboring luciferase fused to 1208 bp of the PRR7 promoter. A genomic fragment containing 2223 bp upstream of the 5 end of PRR7 was then cloned upstream of the PRR7 gene to replace the 1208 bp promoter. Col-0 plants were transformed by the floral dip method. Basta resistant. Homozygous for the transgene; heterozyous for the mutation.
